Early attempts to harness the immune system to fight cancer involved tumor-associated antigens, proteins that are present on the outer surface of tumor cells. Antigens are recognized as “foreign” by circulating immune cells and thereby trigger an immune response. However, many tumor antigens are altered forms of proteins found naturally on the surface of normal cells; in addition, those antigens are not specific to a certain type of tumor but are seen in a variety of cancers.
